이주상 교수님은 2019년 우리 학교 의과대학 기초대학원 교수로 부임하셨고 AI 대학원에도 소속되어 있습니다. 생명 현상을 이론적, 수학적 방법으로 분석하는 연구로 물리학 학위를 받으시고 이후 지금까지 데이터 사이언스를 접목한 암연구를 해오셨습니다. 2021년 최근에도 Cell에 논문을 내는 등, 매우 훌륭한 성과를 보여주고 계십니다. 교수님의 연구실 이름이 차세대 의학 연구실(Next Generation Medicine Lab)이라는 데서 알 수 있는 것처럼 새로운 연구를 지향하고 계시며, 이번 강연에서도 암 환자 개개인에 맞는 진단, 치료법을 제시해주는 것을 목표로 하는 정밀 의학이라는 첨단 분야를 소개해주실 예정입니다. 아래 초록 참고하기 바랍니다. 

Harnessing synthetic lethality to advance precision cancer medicine 

Much of the current focus in cancer research is on studying cancer driver genes. In search for new and effective cancer drugs, this has been translated into searching for ‘actionable’ mutations in these genes, aiming at their therapeutic targeting. However, identifying novel genetic interactions occurring between cancer genes may open new drug treatment opportunities across the whole cancer genome. We focus on the well-known type of genetic interaction, synthetic lethality, describing the relationship between two genes whose combined inactivation is lethal to the cell. I shall describe a new approach for the data-driven identification of these two types of genetic interactions by directly mining patients’ tumor data. Applying it to analyze the Cancer Genome Atlas (TCGA) data, we have identified the first pan-cancer genetic interaction networks shared across many types of cancer, which we then validated via existing and new experimental in vitro and in vivo screens. We find that the identified synthetic lethal interaction network offers an exciting venue for personalized selective anticancer treatments enabling the prediction of patients’ drug response and providing new selective drug target candidates including both targeted and immunotherapy.
