연구 분야: 저희 연구실은 감염성 질환 (패혈증 등) 및 염증성 질환 (자가면역질환 중심)의 병의 발병 기전을 규명하기 위해, 이러한 질환에서 중요한 기능을 담당하는 밝혀지지 않은 새로운 면역세포를 찾아내어 특성을 연구하고 있습니다.
한편, 면역세포의 기능 조절 핵심 G-단백질 결합 수용체 (GPCR) 및 세포내 중요 신호전달효소 (Phospholipase D)에 의한 면역질환 제어 기전을 밝히고자 합니다.
응용 분야: 감염성 및 염증성 질환을 치료할 수 있는 신약 개발을 위해 GPCR 작용약 및 phospholipase D 저해제 등을 이용하고자 합니다.

Research Background |
Leukocytes play essential roles in the regulation of tissue homeostasis and inflammation. Accumulating evidence demonstrate that there is a diverse heterogeneity of each type of leukocyte depending on the characteristics of different tissues or their surrounding environment. We are interested in the heterogeneity of myeloid immune cells, including neutrophils and monocytes. We are trying to identify new myeloid immune cells present in specific tissues or organs at steady state, factors regulating their generation, and their immunological functions. Some myeloid immune cells are infrequently generated under normal conditions, however, they are specifically produced abundantly in extreme pathological conditions. We are also interested in these abnormal myeloid immune cells.

3. GPCRs as important targets to control infectious or inflammatory diseases
We are interested in the development of bioactive molecules that can be used to treat infectious or inflammatory diseases. Our major target diseases include polymicrobial sepsis, asthma, and rheumatoid arthritis. To generate useful target molecules that can be used to develop therapeutic agents against these infectious or inflammatory diseases, we are focusing on several chemoattractant G-protein coupled receptors (GPCRs). Since leukocyte trafficking in vivo is a crucial event that modulate immune and inflammatory responses, and chemotactic GPCRs mediate these responses, the receptors can be considered as important primary targets to control infectious or inflammatory disorders. Currently, we are testing the effects and mechanism of action of chemotactic GPCR ligands against these diseases in experimental animal models.
Sepsis is a serious life-threatening infectious disease. More than 200,000 deaths have been caused annually by sepsis in the US. Since there are no effective therapeutic agents against sepsis at this point, scientists have been trying to identify useful target molecule to control sepsis. We are investigating some chemoattractant GPCRs including formyl peptide receptors and CXCR2 as important targets for the development of therapeutic agents against sepsis.
4. Phospholipase D as a target to control infectious or inflammatory diseases
Phospholipase D (PLD), which hydrolyzes phosphatidylcholine into phosphatidic acid and choline, is a key cellular signaling enzyme. PLD is commonly expressed in various tissues and cells, and is activated by diverse extracellular stimuli including GPCR ligands and growth factors. In addition to normal physiological roles of PLD at steady state, PLD play crucial roles in the regulation of pathological progress of diverse diseases. We are trying to reveal not only functional roles PLD in pathological conditions, but also its mechanism of action.
